For decades, metastatic pancreatic cancer has been one of the most serious diagnoses a doctor can give. Only three out of every hundred patients survive five years, and few live longer than one year. When first treatment fails, options nearly disappear. However, on May 31, a physician from Boston presented remarkable results in Chicago that changed everything. The drug is called daraxonrasib, and it is a pill that roughly doubled how long patients lived. In a global trial called RASolute 302, about 500 patients with advanced pancreatic cancer received either the new drug or standard chemotherapy. Patients on daraxonrasib lived a median of 13.2 months, while those on chemotherapy lived 6.7 months. This represented a 60% reduction in death risk. What makes daraxonrasib different is how it works. Pancreatic cancer is usually driven by mutations in a gene called KRAS. For years, scientists thought KRAS could not be treated with drugs. However, daraxonrasib, developed by Revolution Medicines in California, uses a new approach called molecular glue therapy. It blocks the harmful signaling of mutated KRAS. The results were impressive. Tumors shrank in 31.6% of patients on the drug compared to 11.2% on chemotherapy. Importantly, the new drug had fewer serious side effects than standard treatment. Only 1.2% of patients stopped taking daraxonrasib because of side effects, compared to 11.2% on chemotherapy. The most common problems were rash, mouth sores, nausea, and diarrhea. Revolution Medicines is now seeking approval from the U.S. Food and Drug Administration. Since KRAS mutations appear in many cancers, including lung and colon cancer, doctors believe this drug could help many more patients in the future.
